The railroad company was chartered on January 16, 1836, but financial difficulties delayed construction until 1848.

Its primary routes connected Chicago, Illinois, with New Orleans, Louisiana, and Mobile, Alabama, and thus, the Great Lakes to the Gulf of Mexico.
From there a connection was built north to the Galena and Chicago Union Railroad, finished January 1, 1856. (The west end of the jointly owned line was, and still is, the west bank of the Chicago River.) On March 30, the G&CU and Chicago, Burlington and Quincy Railroad began using it to access the Illinois Central's Central Station.

Galena is the largest city in and the county seat of Jo Daviess County, Illinois, United States. It had a population of 3,308 at the 2020 census. A 581-acre (235 ha) section of the city is listed on the National Register of Historic Places as the Galena Historic District.
